| Abstract | Obesity is becoming a health problem worldwide as has been recognized the World Health Organization has a report a 2020 and May be found in increasing numbers. Obesity is defined as a Body Mass Index (BMI) ≥ 30 kg/m² and a waist circumference greater than 1,020 mm in men and 880 mm in women as the cause of much morbidity which is a result of eating habits and sedentary life styles, can lead to excessive accumulation of fat in various organs may lead to abnormalities in their metabolic processes. It increases the risk of metabolic syndromes, hyperlipidemia, type II diabetes, coronary heart disease, hypertension, osteoarthritis and certain types of cancer include respiratory problems such as obstructive sleep apnea (OSA) and psychological impact. Obesity are related to increase the size of adipocytes (hypertrophy), and differentiation increases the adipocyte number (hyperplasia) which this process is controlled by transcription factor peroxisome proliferator-activated receptor gamma (PPARγ) and cytosine-cytosineadenosine-adenosine-thymidine (CCAAT)/enhancer binding protein-alpha (C/EBPα)which in upregulates genes involved in adipogenesis. The researcher recognized the importance of herbs in the Warayokasan scriptures. Therefore, the objective of this research was to study the inhibitory effect of fat accumulation in 3T3-L1 adipocytes of the mixed extract of Terminalia chebula Retz., Cyperus rotundus L. and Tinosporacrispa L. in the Warayokasan scriptures in a ratio of 1:1:1. Therefore, it was tested with oil red o staining. The herb with the best lipid accumulation inhibitory effect in the water and ethanol extract was TCW, RMW, CRW, TCLW (25.54, 37.50, 46.93, 49.37, respectively), RME, CRE, TCE, TCLE (49.85, 50.46, 51.06, 55.25, respectively), and The effects of treatments on mRNA expression were detected by real-time PCR and showed Treatment with RME, CRE, and TCE significantly reduced the expression levels of PPARγ and C/EBPα proteins, with the decrease becoming more pronounced with increasing drug dose. Compared to controls (p < 0.05), only TCLE showed a difference, significantly increasing the expression levels of PPARγ and C/EBPα with increasing drug dose. Based on test results, it has been found that the herb of interest is a beneficial option, whether used alone or in combination with other extracts. However, further research is needed to develop it into an effective substance and may represent a novel approach for the treatment of obesity that inhibits lipid accumulation in mature adipocytes. |
