| Abstract | This study aimed to evaluate the in vitro anti-diabetic, anti-advanced glycation end products (AGEs), and anti-inflammatory activities of the ethanolic seed extract of Bouea macrophylla Griffith (MPSE) and its major phenolic constituents pentagalloyl glucose (PGG), gallic acid (GA), and ethyl gallate (EG). The total phenolic content (TPC) of MPSE was determined by the Folin–Ciocalteu method and found to be rich in polyphenolic compounds. High-performance liquid chromatography (HPLC) analysis confirmed PGG as the predominant compound in the extract. The results showed that MPSE and PGG exhibited strong α-glucosidase inhibitory activity (IC₅₀ = 6.65 ± 0.48 and 3.02 ± 0.27 µg/mL, respectively), significantly higher than the standard drug acarbose (IC₅₀ = 231.5 ± 3.44 µg/mL, p < 0.05). In contrast, α-amylase inhibition was relatively weak, suggesting selective enzyme inhibition that could reduce postprandial hyperglycemia with minimal gastrointestinal effects. In the anti-glycation assay, MPSE and PGG moderately inhibited AGE formation in a time-dependent manner, particularly during the early incubation stages, while aminoguanidine (AG) showed the highest activity. The inhibition is likely related to the phenolic compounds’ ability to scavenge reactive carbonyl species and prevent protein glycation. In the nitric oxide (NO) inhibition assay, GA exhibited moderate anti-inflammatory activity (IC₅₀ = 30.08 ± 0.28 µg/mL), whereas MPSE, PGG, and EG showed weak or negligible inhibition (IC₅₀ > 100 µg/mL). These results indicate that GA may contribute to the anti-inflammatory potential of MPSE through suppression of iNOS and NF-κB pathways.Overall, this study demonstrates that Bouea macrophylla seed extract, particularly its main phenolic compound PGG, possesses potent in vitro α-glucosidase and anti-glycation activities, supporting its potential as a natural alternative for managing hyperglycemia and preventing diabetes-related complications. |
