| Abstract | Dipterocarpus alatus is a useful medicinal plant due to its ability to produce secondary metabolites. This study aimed to investigate the effects of various concentrations of auxin combined with cytokinin on callus induction derived from shoot tip, leaf, stem and root explants. The experiment was conducted using a completely randomized design (CRD) with three replications. The various explants were cultured on Murashige and Skoog (MS) media containing 1.0 mg/l 2,4-D+0.5 mg/l TDZ (T1), 1.0 mg/l 2,4-D + 0.5 mg/l TDZ + 5.0 mg/l BA (T2) and 2.0 mg/l 2,4-D+0.5 mg/l TDZ + 5.0 mg/l BA (T3). The results showed that T1 media was the optimum media for callus induction obtained from shoot tip and leaf explants, indicated by high surface area, width, length and green callus. In contrast, T3 media favored callus formation derived from stem and root explants, resulting in greatest callus length, surface area, fresh weight and greenish-white callus. These findings suggested that a low level of 2,4-D combined TDZ represents an optimal hormone balance for inducing D. alatus callus obtained from shoot tip and leaf explants. Meanwhile, a higher concentration of 2,4-D combined with TDZ and BA was optimum for callus induction derived from stem and root explants. |
